Only ammo marked "30-06" on the case head- or military ammo for "US Rifle Caliber .30". Do not attempt to fire 30-30, .308, or 7.62 NATO ammo.

The US Military briefly experimented with converting .30-06 to .308 by the addition of a spacer sleeve in the chamber. It was abandoned due to the adapter coming loose during prolonged firing. .32 acp can also be fired through a .30-06 with a cartridge adapter.
The M1 Garand rifle is designed to fire a specific loading called "M2 Ball" ONLY. Anything else at your own risk.

Sounds as if you are looking for an answer to an Armorer's Qual course program- the answer will be found in the TM for the M16 series rifle, and I don't have that. However, anytime a barrel is replaced, the headspace of the weapon must be checked (any rifle, by the way). IMHO, yes, the rifle should be test fired. As to the number of rounds that the TM specifies, you will need to look that up.
