I think most people will not bother either partners of a lesbian couple if they came to know of it, as they will realize that they have little hope in getting through.But still, there might be some nosy guys who might turn out to be very persistent. Specially because men are very fascinated and curious about lesbians. They most appropriate way would be to tell politely "I am sorry to disappoint you but for all you can do, I think she is not going to be flattered the least by what you say.She is more warmer to the attention of other girls". This can be followed by a light peck on your girlfriend's cheek and a soft tug by the waist to guide her away from that person. I think this will be the most appropriate manner because acting rude might attract unwanted wrath from the person, especially if he is biased about the subject of homosexuality.
