Take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to Lexington Avenue-53rd Street.

The Lexington Avenue-53rd Street Station is connected to the 51st Street Station on the 6 train (of the 4-5-6, the dark green line).

Transfer to the Uptown 6, and take the Uptown 6 to 68th Street-Hunter College (at Lexington Avenue). Then walk east to York Avenue.

If it's West 70th Street, you can take the Uptown 2 or 3 express trains (of the 1-2-3, the red line), 2 stops, from 34th Street-Penn Station to 72nd Street (at Broadway).

If it's East 70th Street, you can take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to Lexington Avenue-53rd Street. Transfer is available between this station and the 51st Street Station on the 6 train (of the 4-5-6, the dark green line). Take the Uptown 6 2 stops to 68th Street-Hunter College (at Lexington Avenue).

Depends if you are trying to get to West 77th or East 77th in Manhattan. If you are trying to get west, take the 1 train to 81st St. or the 2 or 3 train to 72nd St. If you are trying to go east, take the 6 train to 77th St.
