Take the Uptown 1 train (of the 1-2-3, the red line) from 34th Street-Penn Station to 50th Street (at Broadway). Then walk north on Broadway to the David Letterman/Ed Sullivan Theatre, on Broadway between 53rd and 54th Streets.

Take the S shuttle train (color grey) west across 42nd Street, from Grand Central Station (East 42nd) to Times Square (West 42nd).

At the Times Square Station, transfer to the Uptown 1train (of the 1-2-3, the red line), one stop, to 50th Street (at Broadway).

The Gershwin Theatre is at 222 West 51st Street between 8th Avenue and Broadway, so walk one block north to 51st Street and half a block west to 222 West 51st between 8th Avenue and Broadway.

So, I just looked it up on Hop Stop (a great app that has all of the NYC subways in it if you want to grab it).

It looks like you can take the D or the B subway lines (15 minutes)... you could also take the Q32 bus (20 minutes), or you could take a taxi (7 minutes), or you could walk (21 minutes).

All of these of course don't take into account waiting time in line or because of crowds. Hop Stop usually does incorporate closings though, so it should be accurate as far as the lines go... if you want to be sure, use the app on the day of your trip... and start out extra early if you know everything will be extra crowded because of an event (like a Papal visit).
