Fort Sumter is nomally taken as the first engagement of the war, with the Confederate artillery firing on the US Army garrison on the island of Fort Sumter in Charleston Harbour on April 12th 1861, even though there were no casualties.

After a number of small skirmishes, the first pitched battle was Bull Run/Manassas (July 21st 1861), a Confederate win.
