The most famous Roman emperor to tolerate Christianity was Constantine I, although the actual official toleration of Christianity was initiated earlier by emperor Gelerius. Constantine supported Christianity and undertook an ambitious programme of Church building, the most notable of which are the Old basilica of St Peter's (in fell in disrepair and was replaced by the current one) the Papal Archbasilica of St John's Lateran (which was Rome's cathedral and the original residence of the popes), the Church of the Holy Sepulchre in Jerusalem, and the Church of the Holy Apostles and the Hagia Eirene in Constantinople.

After Constantine I, the only pagan emperor was Julian the Apostate (reigned 360-363). He tried to restore paganism as the main religion of the empire and to create divisions among the Christians. The next emperor (Jovian, reigned 363-364) re-established Christianity as the favoured religion of the empire. Valentinan I (reigned 364-375) was a Christian, but tolerated paganism.

Next, co-emeprors Gratian, Theodosius I and Velentinian II issued the Edict of Thessalonica (in 380) which established Latin and Greek Christianity (which were the mainstream of Christianity) as the sole legitimate religion of the empire. Later, Latin and Greek Christianity came to be called Catholic and Orthodox respectively. The aim of the edict was to ban other interpretations and sects of Christianity (particularly Arian Christianity which was very popular around the empire) which were branded as heretic. Theodosius I quickly started to persecute the other Christian sects in the eastern part of the empire.

Religious conflict became a conflict between mainstream Christianity and Arianism, particularly in the west, where the young Valentinian II (he was only nine when he was made to sign the Edict of Thessalonica) was an Arian and begun to support Arianism in the west. St Ambrose, the archbishop of Milan, became the centrepiece of opposition to Arianism in the west. Eventually, Arianism lost and disappeared from the religious map.
