Like any name calling, it would really depend on what spirit he meant it in.
If he was joking, it's probably best to let it go for now. If it comes up again, politely tell him if it bothers you. Tell him that you like kidding around with him, but that specific thing bothers you, upsets you, feels hateful, or however it is making you feel.
If he is being mean spirited, you can look for an opportunity to tell him, but it may not help. You might want to talk to your mom or other adult you trust to go to him on your behalf.
If he was being serious, like he really thought you might be, then it is time to sit down and talk to him. Tell him what you are ready to tell him. If you are or you aren't, it is up to you to decide when you want to talk about it. Either way, you need to open up communication and tell him how you feel about him calling you a lesbian. Depending on how you feel, you can even politely tell him you think he might have sexist ideas of gender roles, and that you do not believe the same way he does.
