Something is wrong with your rear door and it is not closing properly in the first place.
Your door lock actuator is broken, it will need to be replaced.
I had a similar problem when I first got my Avenger. My remote was not working, so everytime I locked the door ( while open)the alarm would arm. This is the trick: Lock the door when it's closed, don't lock it when it's open otherwise the alarm will go off. OR You can disarm the alarm by unlocking the trunk with the key. I learned of this from a Dodge dealership. Had issues with the remote and once you lock the car from the inside or out it is armed. I ran into times where i would lock the door after getting in then once i started the car alarm would go off or once locked you cannot open the door from the inside or alarm will go off.
To clarify just a bit. The doors no longer unlock when car is turned off or when using the keyless entry remote. Using the key and the button on the drivers side door still works with out a problem. Also, the doors will still lock when car is turned on and when the doors are locked using keyless entry remote.

We have just bought a Polo with this light on the door. It is a warning that car is safely locked. If you turn lock twice when shutting the door it goes off but car is still locked securely.

How have you locked the car? If you locked it with the key, then the alarm probably isn't armed anyway. If you did manage to lock it with the remote, then unlock with the key. Open the door, and imediately put the key in the ignition and turn the ignition to the run position. The car will recognise the transponder chip in the key, and switch the alarm off if it has started sounding.
The door to go down will be remain locked until you have completed the main body of the game at Realgem Tower.

The key would be favorite.
My first thought would be to check to make sure the "child-proof" setting on the door locks wasn't activated. If that safety feature is activated the doors cannot be opened with the interior door handles.
