Often times when applying for a job, a resume is essential. The potential employer of a job seeker, may ask for a letter of recommendation after a person is hired. If the potential employee has lost his previous job due to downsizing or cut backs, once again the potential employer often asks for a letter of recommendation from the applicants former boss.
If your reference has a prior relationship with the potential employer then the contact would make sense. If not, no.
You will want to format the letter into one paragraph - I broke it up like this due to the personal choice you will need to make in the wording of the letter.------------------------------------------------------DATE OF LETTER HEREDear (Potential Employer),Thank you for the potential job opportunity you have offered to me. A cover letter expressing your interest and summarizing why you would benefit the company. A resume showing your education, job history, and performance of work of interest to the potential employer.
A letter of recommendation provides the potential employer with a way to hear about your strengths from someone who has worked with you. Or, if it is for getting into a college or university, or to earn a scholarship, it helps the reader learn about you from a teacher or another mentor, who can vouch for you.

Short, sweet and to the point is best. Something like "Thank you for the kind words" might be a good start if it's a personal letter. If the letter is from an employer it would be appropriate to start with something like "Thank you for your faith in me" and add something like "I'm excited to get started" and/or "I won't let you down" Of course, finding words that you're comfortable with is what's important. Be sincere.
