It's looking for a Startup Folder. Give it a few minutes to look for it if you could start up normally any other time. If you have more then one operating system on it it may take a few more minutes. If it does finally start up then make sure you have selected a "Start up Disk" in the "Start Up" menu in the System Preferences. If it won't start up then start up with your OS-X CD/DVD and use the Disk Utility to Repair Permissions and Repair Disk.
Plug your flash drive into the USB port on the computer. If a folder does not pop-up immediately, go to your "My Computer" folder and find the USB drive there. Open the folder for the USB drive. Also open the folder the document folder containing the files that you want to put on the flash drive. Drag the documents over to the flash drive folder and a copy will be placed in the folder. Once all of the desired documents have been copied over, you can remove the flash drive from the computer.
Open up the Flash Drive from the desktop, create a new folder out of the flash drive, select the files in the Flash Drive, then drag-and-drop the files into the folder.
If you want to make a new folder go to Computer then click on the button that shows which is your flash drive. Click File or Organize then click on New Folder then name your folder. Next if your picture is on your computer go to the picture then right click on the picture then go to Send to then click on the name of your flash drive. Then go to Computer click on the name of your flash drive then drag the picture to the folder you want it to go. That is all you have to do.
In the Utilities folder within the Applications folder you should find either (or both) a folder called Adobe Installers which will have uninstallers for any Adobe software you have installed or the Adobe Flash Player Install Manager which, when run, will provide an Uninstall button to remove the Flash player from your system.

Once inside your flash drive's folder, follow these steps to organize them: 3a. Right click inside the window of your flash drive's files, hover over New & click Folder. Name the folder (optional), & it's simply drag & drop the files you want from their locations into the new folder. 3b. Repeat to make multiple folders inside your flash drive.

I don't know if this is the answer you are looking for but I used to run from a 32gb flash drive. It loaded a lil slower than from HDD but not bad. I didn't install directly to the flash drive. I copied the folder from the computer to the flash drive. I also copied my lotro folder from my user documents so when I use it on another computer I can copy the user folder (small) to their computer and use my plugins.
