Hundreds of years ago, the owner of a packaging company that sold goods to the United States was knicknamed "Uncle Sam," and on each box the initials "U.S." (for United States, the destination of the goods being shipped), were printed. The workers in the production company didn't know that the "U.S." stood for "United States," and thought they referred to the owner of the plant--"Uncle Sam." Ever since, the name "Uncle Sam" has been synonymous with "United States."
