You would need to delete the partition and create a new one in its place; there is no standardized way of converting NTFS to FAT32. Several tools can be used to do this. cfdisk from the command line, or GParted graphically are probably the best options.

The file size limit of FAT32 formatted hard drive is 4GB, to increase this you would need to format your hard drive and make it use NTFS. Formatting a hard drive erase all the information on the hard drive! The only real limit that would apply is the FAT32 partition size limit, which is about 2 TB. No Flash drive comes close to this size.
