The Properties screen is used for doing this. If you go to My Computer (or Computer on newer ones), you can right-click on a drive letter and select Properties. From there, you go to Tools, and there you find options to defragment, check for errors, and even backup your data.

No! In fact, it's best that you're NOT online while doing a defrag. Anytime defrag hits a part of a hard drive where the information is changing, defrag restarts doing that portion of the drive. While on the Internet, your PC is constantly sending and receiving files and changing what's on the hard drive. Your defrag will take a lot longer.
Yes, you should do Scandisk & repair errors (2 check boxes) first. Go to Start>my Computer. Right click the "D" Drive, choose Properties, then Disk Clean-up which is scan disk. Or, go to Start> Accessories>System Tools, then Scan Disk. After it finishes, you should run Defrag on the "D" Drive. Go to Start> Accessories>System Tools, then Defrag.

It would'nt take forever but yes, normally the bigger your hard drive is, the longer it takes to defrag. It also depends on how bad the hard drive is in need of the defrag.
