Samuel anointed David while he was still a shepherd boy. But he anointed him before Saul was "taken off the throne". By anointing David, Samuel was declaring that he would be the next king, not that he was as that moment the new king (In this case, anointing is not like a coronation). Saul was not even aware that God had told Samuel to anoint David so he let David play music in his court to soothe him while Saul was still on the throne.
