There are three main enzymes involved-
Helicase - This enzyme separates the two parental DNA
DNA Polymerase - This enzyme exists in different forms and each one of them have a specific function in the replication of DNA.
In short, it enhances each strands, adds base pairs and repairs any damage done to the strands during the replication process.
Ligase - This enzyme puts the two stands together after the replication is complete.
In genetics, an enzyme known as DNA polymerase effects the copying of DNA there for it helps in replication, as in MAKING, another enzyme known as DNA helicase is used to break the bonds on the nitrogen bases so the strands can separate, BREAKING bonds.
DNA gyrase is a bacterial enzyme which introduces supercoils into the bacterial DNA, resulting in a highly condensed 3-dimentional struture. it is also known as Topoisomerase. Quinolones and F/Quinolones inhibit this enzyme and thus interfere with bacterial DNA replication. The enzyme is absent in humans.
one of them is heliocase. it 'unzips' the DNA strand. You can always remember this because it's in a popular joke: Q. Why is the enzyme heliocase a lot like a teenage boy? A. They both want to unzip your jeans (genes) !!!!!
One is known as the Leading strand, and the other is known as the Lagging strand.
DNA replication occurs during the S-phase of interphase. Also known as prophase I
Topoisomerase is the enzyme that unwinds the DNA during replication. It binds to the DNA, and separates the double strands and form a replication fork. After which the primer bind to the start site, and DNA polymerase starts DNA synthesis.

Reverse transcriptase is the viral enzyme responsible for making a DNA copy of the RNA genome from a retrovirus. As such, it is technically known as an RNA-dependent DNA polymerase. This is opposite to the usual flow of genetic information from DNA to RNA to protein. This allows a virus with an RNA genome to make copies of itself that can be integrated into the DNA of its host.
