The Anti federalist didn't want to ratify the Constitution because they thought it took to much power away from the states. The Constitution called for a strong central government which meant that most of the powers would be held by the central government opposed to the state's government's. The Anti-Federalist also wanted a Bill of Rights with the Constitution to ensure the people rights.
They believed that the Constitution did not provide for states rights and personal freedoms. The Bill of Rights largely tipped the argument into the federalist's favor. The issue was not fully resolved until after the American Civil War.

The Anti-federalists felt that without specific protections (right to free speech, freedom of religion, right of assembly, etc.) the proposed federal government would threaten the liberties of the people.
The Anti-Federalists opposed the ratification of the constitution, the Federalists were the ones trying to ratify it.
The constitution would empower the national, or federal government, much more than the Articles of Confederation which were in use at the time.
So the Federalists were the ones in favor of a stronger Federal government, and obviously the Anti-Federalists were against it.
