Ferdinand Magellan want to locate a westward route to the Spice Island because the eastward route, was being controlled and monopolized by the Venetians and the Ottomans. Venice was the middleman conducting trade and transportation from the Orient to the Occident that gain them wealth and power (as well as cooperation with the Eastern states and cities) that allowed them to tax heavily on goods in the ships, either docking or passing through Venice. So the goods in the ships that have taken the silk route is very expensive when it came to the markets of different cities in Europe. Urging the European powers like Spain and Portugal to find another route from which they can also monopolize their own trade.
