Charles II granted William Penn the land grant in America because of a debt he owed to Penn's father, who assisted him in regaining his throne. As a young man, Penn had become a believer in the Quaker faith, an act that drew hostilities from other members of the nobility. As a result, he chose to found Pennsylvania as a haven for not only Quakers, but any group facing religious intolerance.
William Penn had been jailed for his religion several times and the King owed his father money. So by getting land in the colonies, he helped himself and Quakers out and the King got rid of some religious troublemakers.
