Edward and his brother were imprisoned in the Tower of London by Richard of Gloucester (Richard III). After that, it is unknown what happened to them. Contrary to popular belief, two children's skeletons found in the tower were not the king and his brother.
Edward IV died and the throne to his son Edward V. Until Edward could take the throne however, Edward IV declared that his brother Richard should watch over Edward V and his brother the Duke of York. Richard III, he is said to have murdered his nephews, King Edward V and Richard, in the year 1483.
Edward V reigned from 9th April 1483 to 22nd June 1483. he disappeared after being sent to the Tower of London in 1483. Richard 111 was blamed for his death, but the circumstances are controversial. It seems that he died before his 15th birthday and was one of the shortest lived monarchs in English history
Edward V son of Edward IV was taken to the tower along with his younger brother. Edward V was to be the King after Edward IVs death, but was declared illegitimate, therefore he could not be king. He and his brother were taken to the tower in fear of their safety after this news. They were under the care of their Uncle Richard Duke of Gloucester later Richard III. The princes were taken to the tower and never heard from again. The rumor is that either Richard III hired someone to smother them or Henry VII had them killed. It has never been proven who actually did it, both had reason to though. Later there were skeletons found in the tower believed to be those of the two princes.

King Edward V of England reigned from 9th April to 22 June 1483. He was deposed by his uncle who became King Richard III, and who is suspected by some as having murdered Edward (or ordering his murder) in The Tower of London.

Richard III
It is unknown how Edward the V died. He was taken to the Tower of London and never seen publicly again. newtest3 ******* It will probably never be known for certain but circumstantial evidence suggests Edward V and his brother Richard Duke of York were murdered in the Tower around July 1483, when Edward was nearly thirteen. Sir Thomas More claims they were smothered and this claim is echoed elsewhere in contemporary sources. In 1674 two skeletons were found buried in the rubble of a staircase leading from the White Tower to a chapel and they were believed to be Edward V and his brother. More recent analysis of the remains revealed a dark reddish stain around the jaw of the larger of the skeletons which, if the bodies are indeed those of Edward V and Richard, supports the notion of suffocation. The how and even the when of King Edward V and his Brother Prince Richard, Duke of York's deaths, they were last seen on the grounds of the Tower of London in mid-1483, it is widely believed they were murdered on the orders of their uncle King Richard III.
