The Colosseum was built in Rome because it was the immense capital of the Roman Empire, and at the time the largest and greatest city in the world. Similar buildings (amphitheatres) were built throughout the empire and other ancient civilizations, but none rivalled the size and majesty of the Colosseum in Rome.
The structure was intended to glorify the Empire and arouse pride in the people of Rome, but more than anything else it was to make the masses happy and therefore easier to govern. Rome needed something to do this and the Emperor Vespasian chose an immense arena where games could be held.
The simple truth is, Rome needed a place to entertain people. Rome may have had a million inhabitants at this time, so it needed to be built on a colossal scale. It was also intended to be Rome's answer to the greatest building of earlier antiquity still standing - the Great Pyramid.
So, the Colosseum benefited by being built on a larger, grander, more innovative scale than ever before and by being in near-constant use for centuries. Rome benefited by pacifying the people and glorifying the Emperor.
