Bigger cells means small surface area. This can badly impair the processes whose efficiency is determined by large surface areas. Eg., respiration, absobtion of nutrients, etc. Hence, cells grow by division rather than growing in size.

There is a limit to the size a cell can be and efficiently consume food and transport waste through the cell membrane. The surface area relative to the volume of the cell decreases as the cell gets larger and finally the amount of waste produced cannot be removed and/or the cell has difficultly ingesting enough energy.

There is also the issue of motility which would be less problematic in an aqueous environment.

A land based giant single-celled animal would not only have difficulty moving it would have a considerable problem maintaining a normal internal environment in areas that were dry and would probably die of dehydration in all but the most humid environs.
