A misfire on a cylinder is caused by either a bad spark plug, bad plug wire, burnt valve, oil fouled plug due to broken piston ring or busted piston, or a blown head gasket. Replace the plugs, then the wires, and if the condition still exists, have a compression test run. If you are loosing coolant then suspect a blown head gasket.
