Marathon running requires much more than using your legs. Most of your core body muscles are actively involved in the act of running. Without a balanced exercise program that includes core (strength) training you will be an unbalanced running and very susceptible to an "over use" running injury.

The answer is simple: Junk in, junk out. No amount of exercise can make up for body that is weak from poor nutrition.

Think of your body as if it were a car. If you build the car out of sub-par parts, it may run for a while, but it will inevitable break down. If you build it from strong steal, and take care that each piece is properly built, it will last you far longer. Calories are the parts, your body is the car.

Your body is literally built out of the food you eat. Just like the car made from scrap metal, it can survive for a while on nutritionally poor food. However, it won't be strong and healthy enough to survive an endurance battle like a marathon. By eating a diet that is comprised mostly of plant foods (vegetables and fruits), low in animal products (meats and dairy) and moderate in complex carbohydrates (whole wheat bread, brown rice, quinoa, etc.) you offer your body a myriad of nutritional building blocks with which it can keep arteries clear, blood cells clean, muscles built and repaired, and generally run like a properly oiled machine. If you eat a crappy diet, or don't give your body enough fruits and vegetables (the most important component of a balanced, healthy diet) your body won't have enough material to properly maintain itself and will begin to fall apart. No amount of exercise can make up for a bad diet.

Big Macs might offer your body a lot of parts to work with, but they're all nearly worthless, and won't build a machine that lasts. Vegetables offer fewer calories, but each one is incredibly useful and strong. Like quality steal, they build a better machine.

Strength is needed to push off the ground with enough force to go a certain speed, and upper body muscle allows you to pump your arms faster, which naturally makes your legs go faster.

How does a marathon runner and the fastest human being differ?

I assume you mean physiologically. A marathon runner has high aerobic endurance, VO2 max, and lactate threshold. This means that they are capable of producing a high level of ATP (muscle energy) for a prolonged period of time utilizing oxygen to feed the muscles. A sprinter has many fast-twich muscle fibers that are trained to produce explosive energy for short periods of time, but without using oxygen, i.e. anaerobically.
