could be a lot of reasons, i suspect that your boot lock central locking motor has seized, does the motor whirr? if it doesn't then i'd spray some wd40 into it, if it does whirr then the mechanism around the lock may have become disconnected or broken.

Took lining off rear tailgate and couldn't find anything wrong, unplugged all the connectors in the tailgate then plugged them all back in and for whatever reason this seemed to reset something and the lock has worked perfectly ever since. Via the remote and via the key.
