First you need to remove the spark plug on the cylinder that is NOT firing. Then get a compression gauge and screw it into the spark plug hole and then move the wire out of the way so it don't get you when you start the engine up. Yes that's right start it up. count to 5 and turn it off and read gauge, The gauge should rear at least 100 LBS or more while engine is running. If it is lower then 100LBS. then check the 1 beside it the same way and read what you got there. There should be no more then a 15 LB. difference. If there is then you have an internal engine problem. A tune up will NOT fix it.
Definition of a tune-up is- A new a/c delco distributor cap and rotor button ONLY. 8 spark plugs, wires, filters and PCV valve A/C delco ONLY.
If you installed all that and you still have the same miss, then you may have a bad fuel injector on that cylinder. That is if the compression test turns out alright. You also should check your engine oil and make sure you don't smell fuel mixed with the oil. I have seen the plastic line that feeds the injector with fuel, crack and cause a miss fire too and mis fuel with the engine oil. Check these things good.
Need to know what year and what engine to help you.
exhaust leak misfire cylinder imbalance
unless a tune up includes replacing the injectors... or could have weak fuel pump, vortecs NEED >56psi at schraeder valve.

If you have a 4 cylinder engine, the number one cylinder is the one closest to the front of the truck. If you have a 6 cylinder, it the on the driver side closest to the front of the truck
Check the spark plug and the wire on the 8th spark cylinder. Corosion, short, or just bad.
