Not likely. I have done this many of times before and plan to do it more and this has never affected my games. You might not want to, just to be on the safe side, but if you don't feel like taking a game out, that's a risk your going to have to take.

The Xbox 360 was designed to perform in either position, the only problem with standing the xbox up is you are risking it falling over. If the xbox 360 is turned on with a game inside and the game disc is spining, if it were to be knocked over the game will be scratched and possibly have permanent damage. I have personally witnessed this on two ocassions and 1 disc was damaged beyond repair
