No. Several Things Go Wrong With Speedometers. Cable Break, Plastic Gear In The Back Of Speedometer Housing, Or In The Housing Of The Transmission. But No It Will Not Hurt The Transmission.

usually it's the speedometer cable that goes bad, you can reach behind the dash and unplug it from the speedometer and with some pliers undo it from the transmission. If you can spin one end and the other does not spin it is bad.

Probably replace the speedometer cable. The cable connects to the side of the transmission, then runs up the bottom of the car to the firewall, then into the back of the instrument cluster. The cable is turned at the bottom by the transmission, and at the top end the cable turns the mechanism of the speedometer.
