If there happens to be a bunch of cranky Asians, this isn't racist If they are labeling all Asians as cranky just because of their accents, this is racist.
Well...
It is well proven that pronounciation gets processed right alongside the actual content of what's being said. Nasal dialects and accents are generally seen as more whiny. Lisping and slurred accents and dialects are often seen as less reliable.
It might be that an Asian accent will appear as cranky to some people.
Now, a sensible person will learn to suppress first impressions like these, and look at what's being said rather than how things are said.
Then it all comes down to exactly what the co-worker said.
Saying that all Asians speaking with an accent sound cranky is a (personal) opinion, not much to be done about that.
Saying that all Asians speaking with an accent are cranky is a statement, that'd be racist.
