It depends on the organism that's being cultured. If it's a bacterial culture you could have the results in a week or two. If it's a fungal culture it could take as much as 8 weeks.
A inoculating loop is used for transfers from culture plates to culture tubes instead of the inoculating needle because the needle could puncture the agar in tube. The loop is much easier as well to get liquid amount into the tube.
If you really wanted to you could definitely determine how many different bacteria are in a mixed culture. You would have to do a lot of testing for specific bacteria though.
