A coarse knob allows quick movement to roughly focus on a specimen. After which, the fine knob is used to finely hone in on the specimen and bring it in to clear focus.
If the fine focus knob is turned backwards the depth of the specimen decreases. When the knob is turned forward the depth of the specimen increases.

Wet mounts are not attached to the slide, it is "floating". If you use an oil immersion lens, you have to put a drop of oil on the slide cover so the light can correctly focus. The oil will cause the slide cover to stick to the end of your Oil immersion lens. When you focus the field of focus will stay the same since the slip cover is stuck to the lens. When you go to change objectives, the cover will travel with your oil immersion lense.

The coarse adjustment (up or down) is used to quickly move closer to or away from the focal point of the specimen. The fine adjustment is then used to fine tune the focus and bring the specimen into clear view.Coarse adjustment means big movements up or down. Fine adjustment means very small movements until the specimen is in focus.
The final focus? Its used for final focusing of the specimen.
First use the coarse focus followed by the fine focus.
