Differential staining is the procedure that are used to distinguish organism based on their staining properties. Use of gram stain divide bacteria into two classes - gram positive which retain crystal violet stain purple colour, gram negative which lose their crystal violet and give pink colour. By this method we can differentiate two different types of bacteria having different cell wall composition that is the reason gram staining used widely as differential staining

Gram staining can be used to classify bacteria into 2 specified groups; Gram positive such as Streptococcus and Staphylococcus and gram negative such as E.coli, Salmonella and Pseudomonas. Gram positive or negative is determined by the cell wall of a bacterium. It is a useful tool in determining bacteria along with other tests such as oxidase, catalase and other chemical reactions which for all effects narrow down the list until you find the family the bacteria is from then the species itself.
