When the B model of the aircraft came out, it was the first single engine fighter that could escort bombers to Berlin and back from England. This meant that the Luftwaffe would have to face both bombers and a fighter that could hold its own against anything that they put up.
The P-51B was introduced in late 1943, and flew several missions over German territory. The 8th Air Force took a break over the winter, not mounting many bombing raids. In February of 1944, the Big Week missions were launched in which for an entire week, heavy raids of 1000 or more bombers were sent to Germany every day and night. This was the first time Mustangs saw major action against the Luftwaffe, and showed that control of the skies would be allied.
In the Pacific, Mustangs were able to escort B-29's to Japan and back following the fall of Iwo Jima (though the Japanese air force was pretty much whittled down by then).
The Mustang was a key contributor to air superiority over Axis territory, and without air superiority, a war is not easy to win.
