Resuspension buffer (solution I) is used for the isolation of plasmid DNA by alkaline lysis method. Bacterial cells, obtained from the culture (liquid culture or colonies grown on agar plate), is resuspended in this buffer. The purpose of this buffer is to provide an optimal starting pH (pH 8.0) and an ideal condition for subsequent lysis.
A fluid thioglycollate is preferably used soon after preparation to minimize the amount of oxygen that is permitted to diffuse towards the bottom of the medium. The longer you wait before using this medium, the more oxygen is allowed to diffuse into it; more oxygen will negatively affect the growth of obligate anaerobes but would positively affect the growth of aerobic organisms.
Hydrogen peroxide is the medium used for the catalase test.
Inspissator for use in the preparation of TB culture medium (TBT-T100IN)
After filtering muddy water, the mud is left behind as a sludge. If the filter is not frequently cleaned of this sludge, the filtering efficiency is reduced, even to stop altogether.

A medium size body of fresh water surrounded by land is called a lake. Lakes can vary in size and shape, and are formed by various natural processes such as tectonic activity, glaciation, or volcanic activity. Lakes are important habitats for aquatic plants and animals, and provide natural resources for human use.

A sculptor could use stone as a medium. Another would use clay as a medium for making pottery. A carver could use wood as a medium.

Supernatant liquid is the upper layer of fluid found after a mixture has been centrifuged. Because of its lower density, the fluid and the components in it have a lesser tendency to migrate to the bottom of a centrifuge tube.
