He was upset because his plan was that Romeo should arrive at the Capulet tomb and take Juliet away at the moment she woke up. But unless he got the letter Friar John was carrying he wouldn't know that, and might do something crazy like buy poison and kill himself. It's odd that it doesn't bother him when Capulet moves the wedding from Thursday to Wednesday even though it means that Juliet will have to spend a whole day cooped up with a bunch of corpses. It's also odd that he didn't remove Juliet from the tomb himself, and disguise her as a nun until Romeo could show up.

At first, the apothecary was not going to sell Romeo the poison because to do so was against the law and punishable by death. But Romeo convinced him by saying that he was so poor and starved that he should just sell it to him anyway. If he didn't sell him the poison, he probably would have died of starvation or disease. Romeo said that either way he might die, so he should sell him to poison to go buy some food. long story short; Romeo uses money to convice him since the apothercaray was a very poor man
