It's not so much a word as a phrase. A star is, well, a star. Back then, Astrology was more popular than it is now and most people would get the idea that the stars controlled your fate. To cross someone meant to oppose them, to get in their way. You can see where we get the idea of someone "crossing your path". That originally meant that they were getting in your way and preventing you from passing. They were "a-cross" your way.

So it would appear that the stars, which to the astrologically-minded represented fate or cosmic forces, were getting in the way of these lovers. Which is just what happens in the play. They are terribly unlucky.
