You can't. Rome is a one-time locale, and cannot be accessed outside of sequence 14. This is due (A) to the fact that it only appears in one sequence, and (B) because it was not fully created in assassins creed 2. It was simply a linear path that leads to the end. Not the open-world city that appears in Brotherhood

Assassins Creed Brotherhood because in those games you can free roam and you can stil shoot like with a crossbow (i think) and you've got new abilities like riding a horse IN a city and you can man a canon. So my choice Assassins Creed Brotherhood but if you don't know the storys of the other games then its hard to understand youre choice.

They come as a separate class. However, the equipment (secondary blade) for them are dropped from monsters or can be purchased by NPCs probably after or right before the class comes out.

It is believed that they used short daggers or knives, sometimes poisoned. But really they wear nothing like Altair although I wish they were. to reach their target they dressed as surveillance or sometimes infiltrated organisation's They did not always kill, but left warnings e.g. an assassin crept into Saladin's guarded tent and left a dagger in a cake next to his bed. They were much more stealthy than it is possible to be in Assassins Creed, although from what I hear things may change for the sequel.
