In ACB, there are 10 glyphs. The glyphs are positioned on 10 random buildings in Roma, and are marked with a red eyeball logo in the database.

You can only see Glyphs using EAGLE VISION, and when you find them you climb into them to activate them.

Once you enter a Glyph you will be asked a series of puzzles - stupid and often complex ones - go on youtube for answers if you need to.

When you have "solved" all 10 Glyphs, Subject 16 appears to you in a interactive message to explain what's going on. It only creates more questions really tbh.

if by secret location you mean the tombs, then the answer is that if you finish all of the tombs (to finish a tomb, you have to go in one and get to the other end, where you will find a grave casket thingy, you have to open it and take the artifact that's inside it). if you collect all the artifacts, you will get Altiars Armor.

You can't. Rome is a one-time locale, and cannot be accessed outside of sequence 14. This is due (A) to the fact that it only appears in one sequence, and (B) because it was not fully created in assassins creed 2. It was simply a linear path that leads to the end. Not the open-world city that appears in Brotherhood

yes and no it still features desmond but there is a new assassin ezio auditore da firenze and also there is altair in part of the game look out for assassins creed brotherhood

You can't its just there for graphical purposes. There are however, "dips" around the wall but there is no way in entering.
