It's the Roanoke Colony you are talking about, actually. Same place, different people. Jamestown, Virginia, was the first permanent English settlement in what is now the United States of America.

Before the Jamestown people settled there, there had been a colony living there called the Roanoke colony. Nobody knows what happened to the Roanoke settlers. It seems they just vanished without a trace one day. No bodies were found, even. All of the houses had been deliberately dismantled, so apparently they did not leave in a hurry. The only clue ever found were two carvings in two different tree trunks, which read, "Croatoan," and "Cro," respectively.

No one knows for sure what all the Indians' names were because everyone in Jamestown just called them, "Powhatan" because that was the name of the tribe that lived in the area at that time... The population of the Jamestown settlement varied in the 1600's due to several factors, specifically disease, starvation, and poor relations with native Americans. Originally 107 colonists came over to settle the colony. More colonists came over, bringing the total up to 500... but due to poor location and other factors (swampland that had stagnant water, which led to poor health) only 61 of the original settlers survived the winter of 1609-1610, known as "the starving time".

After much needed supplies and more settlers arrived from Lord De La Warr (Delaware later named after him in honor of saving Jamestown), the population stabilized and steadily grew. But in 1622, when the population was about 1,000, a surprise attack by the Powhatan tribe slaughtered around a third of the population, and again in 1644.

The Separatists were religious groups of people who wanted to remain separate from the other groups in the English colony. With their decision to remain separated, they eventually departed England and headed toward America where they had expected to live together and practice their own ways of living with no interference. They spent sixty five days in the Atlantic Ocean throughout their journey. In 1620, they travelled on the Mayflower with the intention of landing in Jamestown. Eventually they made it to the north of Cape Cod. Although the Separatists' ship had been blown off course, it is possible that this was part of their original plan. They may have intended to travel to a place where they would be left alone to live the way that they wanted.

Roanoke Colony, founded by Sir Walter Raleigh, preceded Jamestown but it disappeared. Had it survived it would have been the first English colony.

In 1619, the Virginia Company sent women to the Jamestown settlement to help establish families and promote a more stable community. The arrival of these women aimed to encourage the settlers to put down roots and create a more sustainable colony, as the initial settlement had primarily consisted of men. This initiative was part of a broader strategy to attract more settlers and enhance the colony's viability.
