The first two are in present day, so the years that they came out.
The third is based 10 years in the furture (in the majority of the game).
the fourth is based 5 years in the past. That's all that's out in England at the moment but the following two games are based just after the fourth.
I do not think Professor Layton is based on any particular year, we have to remember that these games are all a fiction and just try to create an era to set the mood.

Professor Layton and the Specter's Flute is set three years before Layton and Luke have their adventure in St. Mystere, so Flora is still living there with her father. Flora is replaced by Emmy, Layton's new assistant. Professor Layton and the Unwound Future was her last appearance in the video game series.
In Professor Layton, Luke is Professor Layton's apprentice/assistant. He goes with Layton everywhere. His father [Clark Triton] is collage friends with Layton so that's how they met. Also Luke is professor Layton's friend (professor layton says so in the lost future).
