It is only available online at minecraft.net, as of yet you can not buy it at a "brick & mortar" store or an online store.
In order to purchase it you will need an e-mail address and pick a user name and password to register and they will send you a conformation e-mail for you to start the download. It will download a small file then it will download the full client, it only takes a few minutes to download the main game, less then 5 minutes with my low end DSL.
It should be around $26.00 US, they take major credit cards and have just enabled PayPal.
