Until 1664 " New York" was a the Dutch colony ( 1625-1626) New Amsterdam settled by a Dutch trading company. So, in 1624 the only people living there were the native Americans that had been located there for thousands of years.

The approximate population in what was then called New York City was 3,000. The phrase of "what was then called " is used because for the most part the city was the island of Manhattan. Later on NYC included a larger area. Present day NYC includes Brooklyn, Queens, Bronx and Staten Island as well as Manhattan.
