If you're referring to the filing of a claim for public domain land under the Homestead Act of 1862, a prospective homesteader would file a patent request with the US Department of the Interior's Bureau of Land Management (BLM), which had land offices in every county where there was public domain land. Once the conditions of the homestead patent were met (building of a residence and development of the land for a specific period of time, usually 5 years) the patent would be deemed to be "perfected" and a deed would be issued.
HOWEVER, homesteading was closed off in 1972 throughout the united states except for certain portions of Alaska, and then only by permission of BLM under certain specific conditions.

Then they had to live on the land for five years and make improvements to the land by builiding a 12x14 dwelling and planting crops. After five years, the homesteader could file for the deed of title by submitting proof of having lived on the land and having made the improvements to a local land office.
