In all likelihood, no. The Avengers: Earth's Mightiest Heroes focuses on the early Lee/Kirby stories which featured the popular Avengers characters: Iron Man, Captain America, and Thor. This show also features characters who appeared in The Avengers comics for many, many years but hardly ever appeared in a cartoon series.
Frankly I'm perfectly fine with Spider-Man not appearing in The Avengers: Earth's Mightiest Heroes. He's starred in many Cartoons before and him not appearing gives other cool Marvel heroes who aren't in cartoons very often a chance to shine for a change.

In a fight between the Avengers and Sandman, Hawkeye would completely be useless. He might even get injured. You would need Iron Man and Spider-Man to work together. Iron Man can distract Sandman from the sky while Spider-Man quickly gains access to water and solidifies Sandman. Iron Man can then use explosive charges to wrap things up and win the battle. Go, Avengers!
Justice League hands down. The Avengers, despite their storied crack-down on crime across the world, simply do not compare to the Justice League, who have ridden the universe of evil, injustice, and the Black Plague. In a battle of wits, the Justice League would besmirch the Avengers' cunning with ease. In an egg-throwing contest, the Justice League would use their super powers to hatch a mighty chicken who would fly from player to player. The Avengers, in their attempts to best the Justice League, would errantly hatch a rooster, and would thereby lose. In competitive civil conversation, the Justice League would be the center of attention, whilst the Avengers would be the butt of all jokes.
