May be nothing more than worn out plugs or plug wires. The car is 9 years old. Replace the plugs and plug wires. If the miss is still there, it may be clogged injectors or burnt valve. Pour a can of Seafoam or Techron fuel injector cleaner in a full tank of fuel to possibly unclog the injectors. I am assuming you are not loosing coolant. If you are you may have a blown head gasket.
my Lexus rx300 misfired as well and i took it to the mechanic and it was the fuel injectors that where dirty because of bad gas

Number 2 cylinder is misfiring.
To remove the ignition cylinder on a 2000 Lexus RX300, first remove the vanity engine cover by unscrewing it. Disconnect the wiring connector by pushing the clip that holds it in place. Finally, remove the bolts that hold the ignition cylinder.
Looking at the engine while standing at the front of the vehicle. The #1 cylinder is the back left corner.
