In the use-dilution test a chemical is evaluated by its ability to kill 106 and 108 dried clostridium sporongenes or bacillus subtilis endospores. why is this considered a stringent test?

Chemicals do not readily penetrate bacterial endospores. The number of endospores used is higher than would be expected in normal use of disinfectant. It uses a measure of determining the effectiveness of a disinfectant using serial dilutions.

Why were you instructed to discard the toothpick in disninfectant?

After using a toothpick, bacteria from one's mouth may reside on it. Because these bacteria can cause illness and contaminate a laboratory, these bacteria must be destroyed. To accomplish this, the toothpick is discarded in a disinfectant, which kills the bacteria.

Why would endospores not be present in a 12 hour batch culture but would be present in 14 day batch culture?

Endospores only form under unfavorable conditions that threaten the survival of the bacteria (and only some bacteria are capable of producing endospores). Also, the process of endospore formation requires 8-10 hours. Assumably, the 12 hour culture would still have an abundant supply of nutrients and therefore would not require the production of endospores to survive; the 14 day culture would have a much higher number of organisms which had been using up the supply of nutrients, therefore creating unfavorable conditions and requiring the formation of endospores for survival.

What do you do if you drop milk on your book?

Blot it up as quickly as you can, then sponge any residue off the pages and binding with a damp cloth. You can use a very light spray of disinfectant to prevent bacterial growth on the exterior of the book, but it may progress within the binding. If it is valuable, there are professionals who clean books using mild chemicals.
