I'm sure you could take the bus, but you'd probably have to transfer at least once. I don't know. I've lived in Manhattan all my life and I still can't figure out how to take the bus, except for the routes in my neighborhood, and some of the crosstown routes. The maps and schedules on the MTA's website are totally incomprehensible.
I would just take the subway, unless you can't because you are walking disabled. The majority of New York City's subway stations are not wheelchair accessible.
Take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to Lexington Avenue-53rd Street.
Then walk 4 blocks south to 49th Street and 1 block west to Park Avenue.

First, are you absolutely sure that you're going to Park Avenue, not Park Avenue South? Because there is a 250 Park Avenue, and also a 250 Park Avenue South. If you're sure it's regular Park Avenue you're going to, then you are headed to East 47th Street. Getting there is pretty straightforward. When you exit Penn Station, you will come out either at the West entrance or the East entrance. The West entrance is between West 33rd and West 34th, on 8th Avenue, and the East entrance is between West 33rd and West 34th on 7th Avenue. Walk east on 33rd or 34th until you hit Park Avenue (three to four avenue blocks). At Park Avenue, turn left, and walk north to East 47th Street. The address -- 250 Park Avenue -- is between East 46th and East 47th, but closer to 47th. If it's Park Avenue South you're looking for, then the directions are almost the same, except when you hit Park Avenue, you need to turn right, and walk south until you get to East 20th Street. Walking directions from Grand Central Station to the Winter Garden Theater?
The Winter Garden Theatre is on 7th Avenue, between 49th and 50th Streets. For some reason, the address of the theatre is 1634 Broadway, when in fact it is on 7th Avenue, not Broadway. I entered the address "1634 Broadway" on Google Maps, and the location it showed was on 7th Avenue, not Broadway. I even zoomed into Street View, and indeed, the theatre is right there on 7th Avenue. I'm looking at a picture of it right now, and it is very obviously on 7th Avenue. I have no idea why the address would be 1634 Broadway when the theatre is not on Broadway, but I thought it worth mentioning to avoid confusion. To get there from Grand Central, walk west on 42nd Street until you get to 7th Avenue. Turn right, and walk north on 7th Avenue to 49th/50th Streets. Grand Central Station is on the north side of 42nd Street, and traffic on the north side of 42nd Street flows west. So, when you exit Grand Central, it should be very easy to figure out which way is west: just follow the direction that traffic is going on the side of the street that you are on.
