Technically, Yuuki was a vampire all along, just that her memories were buried deep into the back of her mind, so it covered it up and made her believe she was human. Her vampire past is awakened when she starts to remember it, and is finally bitten by Kaname Kuran in chapter 35 of the manga, or episode 7 on Vampire Knight Guilty, which awakens her Pureblood side and brings her to realise that Kaname is both her brother and fiance.
