You must be noticed by a supervisor, and if the supervisor sees fit, will recommend you to become a Premier Answerer. Your contributions will then be reviewed by a panel.(Note: A Supervisor cannot become a Premier Answerer because the Premier Answerer badge is a regular contributor badge, not a Supervisor-applicable badge. However Premier Answerers may become Supervisors, although their Premier Answerer status will be removed.)

When a supervisor finds a normal contributor who they think have qualified for the premier answerer badge the supervisor can recommend them via a link that only supervisors have. A supervisor will go through the user's contributions to make sure that they have had quality and well thought out contributions. Please do not ask to become a premier answerer, you must be nominated by a supervisor.

A Premier Answerer is completely different to a supervisor; these users are selected due to the quality of their answers, and not their general contributions towards the site. However quite a few Premier Answerers are also offered a supervisor position, but turn it down. You cannot be both a Premier as well as a supervisor.(See the Related questions.)
