Yesy, it can jump a tooth if worn or be installed without the timing marks lined up properly. This is probably an interferance engine so check for internal damage if it's not running or died while driving

If you're referring to cam timing you don't. That's set when the belt is installed by matching up timing marks on the cam drive wheels and the crankshaft.
