In my opinion you should start with the dog crate next to you, and start moving it away from you little by little.
I hope the Mother is coming with the one week old pups who should not be away from their Dam. Just put them in a nice sized crate with lots of towels and maybe a couple of warm warm (not hot) hot water bottles in with them, wrapped in towels so the pups cannot accidentally burn themselves by lying right up against the hot water bottle, need a towel wrapped around it. Put the Mom in another crate right next to them. They all should be fine for a 40 mile trip. You just need to keep the pups warm for the trip. Use an appropriate size crate to put them in.
